I also don't think her act of murder will be anything to do with her being secretly evil. It would go against everything we've seen of her so far, and would mean the little girl from Silence in the Library is stuck for eternity with a psychopath.
My guess is that she'll be forced to make an impossible decision at some point, and as we saw in the finale, she's not as averse to killing as the Doctor.
River can't be the Rani, nor can she kill the Doctor. That would contradict the Library episodes where she dies as River and states that the Doctor was with her the night before she went there.
I reckon she kills Rory. That would have an effect on both Amy and the Doctor and change things.
So, she managed to become a Professor (Presumably of Archaeology.) whilst in jail?
It has been confirmed that it will be impossible for the Rani to return in the revived series as the character's creators have not agreed to transfer their copyright of the character to any of the new producers, though it may happen one day where the copyright will be taken.
