Return of the Living Dead - When Burt knocks tarman's head off with the baseball bat
Death Proof - The whole chase scene at the end when the girls are chasing Kurt Russell's character. I love how the tables turned in the that scene.

FACE / OFF At the airstrip when his cloak blows open and the Gold Guns are revealed along with the money clip and stuff.

Spider-Man - When Peter raises his head and throws the trident back at the Green Goblin
Spider-Man 2 - The entire "I'm back!" scene up to where Peter starts to fall
The Amazing Spider-Man - Crane scene (the theme music gets me every time!)
TDK - Batman taking out the SWAT team
DKR - Batman's flaming symbol (Music is awesome here too), Bruce escaping the pit.

There are others, but for me, the music helps make these moments. Good superhero themes played at the right time are uber-effective. These moments to me are some of the best examples of " YES!" moments.

Such a classic "YES" moment. "Smile you son of a......" opcorn

I will never forget that moment sitting in the theater in 1975 at 10 years old...at that time we didn't know how it would end....didn't know Hooper would still be alive....there was such a cheer in theater and a standing ovation...and people stood and clapped through the credits...I have never seen anything like it since.

Plenty of the ones mentioned before. Along with:

The Firefly pilot episode ("Serenity") at the end when Mal shoots the Alliance spy without breaking stride.

Pegasus showing up to rescue Galactica (right after the equally cheerable "Adama maneuver.")

And, going back a bit, Willow's activation of ALL the slayers in the "Buffy: TVS" finale.

For me, watching Leverage gives a lot of "YES!!!" moments, but nothing compares to this scene. Eliot Spencer, the team's resident bruiser (and described by Word of God as the equivalent of Batman), does not like guns, and has done his job without the use of them from day one... until now.

Cue the epic badassery when Eliot has no choice but to use guns.

District 9 when Sharlto Copely jumps inside the prawn mechsuit and starts tearing up the mercenaries.

Game of Thrones when you find out Khaleesi spoke valeyrian the entire time and says Dracaris.

Aliens when Hicks shoves the shotgun in the xeno's mouth and shouts "Eat this!"

When Samuel Jackson as Nick Fury mentions "the Avengers Initiative" at the end of Iron Man.

The Mist, when Toby Jones shoots Marcia Gay Harden, I cheer every time I watch the scene.

Optimus Prime blowing through the entire Decepticon line in the opening of Transformers : The Movie.

Death's intro and Death cutting a deal with Dean in season 5 of Supernatural.

Pretty much any and all scenes with Death. That dude has a screen presence.

The elevator "ding" in Cabin in the Woods.

Dredd... pretty much the whole movie.

Guy with the hair in Dredd. If you've seen it, you know the guy.

Superman wailing on Zod in Man of Steel. "YOU THINK YOU CAN THREATEN MY MOTHER?!?!?!?!"

Jor El kicking ass in the opening to MOS.

The Mark 1 booting up and lights coming on in the beginning of Iron Man.

The Mark 2 pre flight check in Iron Man.

Hulk learning Loki a lesson he will never forget in The Avengers.

13! 13! 13! 13! in Rome.

So many more, but those are the ones from recent memory.
